3. Experimental method

　　(1) Screening culture
 
　　①Neutral medium
 
Potassium salt, ammonium salt, magnesium salt, iron salt, calcium salt, sodium salt, phosphate, etc. were sterilized to prepare neutral liquid culture medium; Adding a certain amount of beef paste, peptone and agar powder, and sterilizing to obtain neutral solid medium.
 
　　②Acid medium
 
　　 Use potassium salt, ammonium salt, magnesium salt, iron salt, calcium salt, sodium salt, phosphate to prepare a solution, titrate with acid and alkali, and prepare an acidic liquid culture medium after sterilization;
 
　　 Adding a certain amount of beef paste, peptone and agar powder, and sterilizing to obtain acidic medium.
 
　　(2) Purification and segregation
 
After the strain is activated, it is dissolved in sterile water, shaken and cultured at the corresponding temperature and rotating speed, the supernatant is continuously taken for enrichment culture, and the dominant strain is distributed, then the nutrient solution culture medium is prepared, and the single colony is cultured, and the colony characteristics such as colony size, color and transparency are observed. Then, the strains were separated and purified for many times.
 
　　(3) Production culture conditions
 
　　① Calculating colony density by blood cell counting plate method and drawing bacterial growth curve;
 
　　②By changing the culture temperature, count the colonies and get the best production temperature;
 
　　③By adjusting the pH value of the culture medium, count the colonies and get the best pH value.
